15N in amino acids

Samples are derivatized with TMS according to Gullberg et al 2004. The resulting TMS derivatives are analyzed on a Gas Chromatograph - Isotope Ratio Mass Spectrometer - Mass Spectrometer (GC-IRMS-MS). The instrument setup consists of a gas chromatograph (Trace GC Ultra) connected to both a continuous flow isotope ratio mass spectrometer (DeltaV) through GC IsoLink and an organic mass spectrometer (ISQ LT), all from Thermo Fisher Scientific, Bremen, Germany. Isotopic signatures of the samples are calibrated using inhouse laboratory amino acid standards, which have been standardised against four certified standards:IAEA-600, IAEA-N-2, USGS40, and USGS41

J Gullberg, P Jonsson, A Nordström, M Sjöström, T Moritz, Analytical biochemistry 331 (2004), 283-295: Design of experiments: an efficient strategy to identify factors influencing extraction and derivatization of Arabidopsis thaliana samples in metabolomic studies with gas chromatography/mass spectrometry
