Berg, Cecilia; PhD, Associate professor
Dept. Environmental Toxicology, Uppsala University, Uppsala, Sweden.
Current research activities: The main focus of my research is sex differentiation and reproductive system development and how these processes are affected by exposure to chemicals. Transient exposure to chemicals during critical periods of embryo-foetal development can result in reproductive disorders and impaired fertility as the animal become adult. Using the frog as a model we investigate endocrine disruption, developmental and reproductive toxicity using methods that range over several levels of biological organisation including molecular, physiological, and behavioural. Current research activities:
The research of my group is focused on effects of environmental contaminants on the development of avian embryos. Embryos are exposed in ovo early in development via injection into the yolk sac. Effects of various endocrine disrupters on development of the reproductive organs and on the male reproductive behaviour in quail following embryonic exposure are presently studied. Expression of estrogen receptor α and β in brain and reproductive organs in quail embryos at various developmental stages is studied using PCR and in situ hybridisation. In another project the response of fish gills to dioxin-like compounds (Ah receptor agonists) in the water is studied and a method for monitoring such compounds in recipients is developed.

Reproductive herd health for dairy cows: The main goal for the dairy farmer is to get one living calf from each cow per year. Very few farmers succeed due to reproductive losses e.g. uterine disorders, ovarian disorders, embryonic death, abortions and stillbirths. My research area is focused on factors causing reproductive losses in dairy herds. The ultimate goal is to define and risk factors for reproductive disturbances and by e.g. reproductive herd health programs inhibit the risk factors and prevent reproductive diseases.

Mainly interested in prostaglandin biosynthesis during different physiological (luteolysis, parturition, uterine involution) or pathological (endotoxaemia, fasting, stress, abortion, premature parturition and stillbirth, postpartum endometritis) events. These studies are done on a comparative basis including several domestic species. A strong focus today in the research activities towards exaggerated sexual behaviour and endometritis in mares. Intensive studies concerning use of anti-inflammatory drugs (NSAID:s) to control prostaglandin biosynthesis in eg. luteolysis and inflammatory processes. These studies include clinical changes, prostaglandin formation and pharmacokinetic studies of the drugs. Have a long tradition to analyse prostaglandin metabolites using RIA:s.

Reproduction and infections: Bacterial infections in reproductive organs such as the uterus and mammary gland are major obstacles in animal husbandry. Very often these infections are caused by commensal bacteria why we have focused on the body’s defence mechanisms, specifically the immune system. In the mare we study the uterus and in the sow the mammary gland. The aim of these studies are to provide a basis for better preventive and therapeutic measures for endometritis and mastitis. Also, currently we perform epidemiological studies of the bacterial zoonosis leptospiros among pigs in Vietnam.
Environmental reproductive toxicity: This is a new research field in which we study the effect of low doses of compounds present in the environment on the reproductive functions of pigs.

We perform baseline studies on perch and roach in Swedish waters with a focus on gonad morphology, this in order to describe the cause to inadequate gonad maturation and intersex. In addition we are focusing on reproduction disorders in several Baltic Sea fish species, i.e. cod and salmon (M74 syndrome). Another important task is to develop test protocols for zebrafish including short-term and confirmatory tests which can be used to develop OECD guidelines. Recently we have started to work with amphibians, both native and exotic species. Major groups of chemicals which we are interested in includes musks, brominated flame retardants, PCBs and PAHs. We are also working with infectious diseases in salmonids (renibacteriosis and flavobacteriosis). For more information you can look at www.pat.slu.se (Aquatic ecotoxicology).

Endometrial angiogenesis (the formation of new blood vessels) is important for fertility and bleeding control. We study the expression of regulators of angiogenesis and blood vessel function in in serum, peritoneal fluid and tissue biopsies from normal and dysfunctional endometrium, endometriotic lesions and peritoneum.
Pre-eclampsia (PE) is studied in a new rat model developed by us and in humans. Oxidative stress is believed to be involved in the pathogenesis of PE and it has been shown that antioxidants like vitamin E can reduce the incidence of the disease. inhibitors of. The rat model is used to test hypothesis about the pathogenesis to PE and how the development of PE can be blocked.
We also study the relationship between uterine leiomyomas and endocrine disruptors (PCB, DDT and others, and, their metabolites) in the baltic seal and humans. We have found that leiomyomas occur quite frequently in seals in the baltic sea but is rare in the same species in the Atlantic sea. In vitro studies have pointed out some of the metabolites as strong stimulators of proliferation on uterine myocytes from seal. Human cells have not yet been tested.
Methods used are cell culturing, cell proliferation assays, immunohistochemistry, western blotting, etc.

Prostasomes are membrane-surrounded microvesicles (mean diameter 150 nm, range 40-490 nm) secreted from epithelial cells of the prostate gland. Prostasomes constitute an important part of the prostatic fluid and thereby being a prominent component of the seminal fluid. They promote sperm forward motility and protect sperm cells from attacks from the female immune system and exert complement system-inhibitory activities thus supporting the fertilization process. We demonstrated nucleic acid presence in prostasomes (both RNA and DNA) and the prostasomal membrane exhibits an unusual composition quantitatively dominated by saturated phospholipids with predominance of sphingomyelin. Prostasome-like microvesicles have been observed in other species including, ram, dog, stallion and boar. Research about the prostasomal microvesicles involves important aspects of the reproduction process and we now highlight the characterization of prostasomes in other mammalian species.

The overall aims of my research is to investigate how reproductive hormones influence mood and anxiety in women, how reproductive events affect mood disorders in women, and vice versa, how mood disorders affect reproduction in men and women. My research investigate the interactions between ovarian steroids and mood/anxiety during various aspects of a woman’s fertile life such as young women with adverse mood effects from combined oral contraceptives, pregnant and postpartal women, women with severe premenstrual symptoms, women with postpartum depression, postmenopausal women on estradiol treatment and involuntary childless couples. The study designs span from population-based studies on postpartum depression and its’ genetic component to human experimental studies on anxiety/fear and to neuroimaging studies where functional magnetic resonance imaging is used to evaluate activation of parts of the central nervous system known to be involved in anxiety and/or fear. Furthermore, part of the proposed study is also aiming at elucidating possible harmful effects of progesterone metabolites.

Current research activities:
The research work is related to milking with the focus on the biology of the dairy cow. It involves both physiological and behavioural responses of the cow during milking as well as udder health. Within our research group we have indicated the importance of the management related to milking, since it influences both the physiology and behaviour of the dairy cow. The effects have rather important consequences for the cow. At present much work is also done in relation to automatic milking systems, where the studies focus on milk quality and uder health. Furthermore the effect of early interaction between the cow and calf is studied with regard to the calf´s behaviour, feed intake and growth and the cow´s milk production and udder health.
We also work with milk production in water buffaloes and with milking and milk quality in developing countries such as Vietnam and Burkina Faso.
