Prior knowledge

The enrolled doctoral student should have basic knowledge in molecular biology, preferably have taken molecular biology related courses or practices.

Objectives

After completing the course, the student should be able to:

• Understand the application and limitation of different genotyping methods available for plant mutagenesis screening and genome editing;

• Design genotyping analysis (focused on the methods introduced in the course) to screen mutation lines in their own research project;

• Plan and conduct genotyping analysis independently and correctly; provide good supervision for other students in the future,

Content

To meet the high demand of genotyping tasks in doctoral research projects like mutagenesis breeding (eg.Targeting Induced Local Lesions in Genomes, TILLING) and genome editing (eg. CRISPR/cas9) especially in non-model plants, the course will:

• introduce the principle and the application of advanced genotyping techniques eg. high-resolution-melting PCR (HRM-PCR), high resolution fragment analysis (HRFA) and capillary electrophoresis etc.

• provide hand-on training on the general good practices for genotyping analysis in molecular lab and the specific equipment(s) and available genotyping platform in the Department of Plant Breeding, Alnarp, SLU.

Pedagogical form:

Teaching form will include 3 parts: Self-study, lectures and lab practices

1. Self-study on literatures about the principles of different genotyping methods and their general applications. Some important literatures or references will be uploaded to the course website prior to course start.

2. Lectures will be given focusing on the specific applications of genotyping techniques that are useful in non-model plant research; QA session for part 1 will be integrated to the lectures.

3. Lab practices will be the main focus of the course which is to help the students get familiar with the current protocols and facilities related to genotyping analysis in the Plant Breeding Department, SLU, Alnarp.



Examination:

Students will pass the course and awarded to the full course credits if they have attended all the course activities.
