Applied Plant Biotechnology

The course includes the following aspects:

- theory of tissue culture, in particular in vitro shoot and root culture, genetic transformation, gene cloning and various techniques in molecular biology.

- practices in establishment of in vitro cultures, meristem culture and in vitro rooting, genetic transformation via Agrobacterium-mediated gene transfer, DNA- and RAN-isolation, Southern blot and qRT-PCR analyses

- demonstrations of transformation methods such as particle bombardment or electroporation

- project design or case study focusing on specific problems, e.g. rooting capacity, disease resistance and quality aspects to illustrate potential solutions that biotechnology can provide.

-seminars.

- excursions to institutes or companies working with plant material and biotechnology.



Teaching forms include regular lectures, a number of laboratory practices, study visits, literature study, case study or project design, seminars and discussions.

Enrolled PhD who have already earned 120 HEC in Biology including plant physiology and genetics.

Mål

The changing climate and an increasing world population require more food and feed production in a sustainable way. Plant breeding has a crucial role in producing healthy and environmental friendly food and feed by producing new cultivars that can better adapt to a changing climate. Plant biotechnology including tissue culture and gene technology is an important modern breeding tool in producing such new cultivars (referred as biotech crops hereafter). The overall objective of this course is to provide the PhD students a better understanding of biotechnology application in agriculture and its potential use in the future, the important tissue culture and molecular methods used for producing and analysing biotech crops, and the current regulations about biotech crops within Europe.



On completion of the course, the students will be able to

- perform tissue culture work independently

- choose the plant transformation methods depending on purpose and perform plant transformation independently

- independently choose and use the most commonly used molecular methods in their own studies.

- get more familiar with specific regulations about biotech crops and with the biotechnological companies in Sweden

Examinationsformer och fordringar för godkänd kurs

The marking scale is pass or failed. The examination consists of a written report on designed project (or case study) and oral presentations (literature study, laboratory practices and project design). To pass the course, one needs to participate in all compulsory activities (project design, all seminars, at least two lab practices), to have approved written report and oral presentations.
